The central bus station has lockers that you can store your luggage in from anywhere between 1hr - 24hrs. The rates are inexpensive with the 1 - 2hrs fee approx $4. They are coin operated, so be sure to have some coins with you.

If you are going to catch a coach tour to Victor Harbour or Kangaroo Island, this is the place you'll need to go to early in the morning. Overnight coaches from Sydney, Melbourne, Brisbane and Perth also arrive here early in the morning and depart at night.
